Engineering a Metathesis-Catalyzing Artificial Metalloenzyme Based on HaloTag

Artificial metalloenzymes (ArMs) are created by embedding a synthetic metal catalyst into protein scaffold. ArMs have the potential to merge catalytic advantages of natural enzymes with reaction scope catalysts. The choice scaffold is utmost importance tune activity ArM. Herein, we show repurposing HaloTag, self-labeling widely used in chemical biology, create an ArM for metathesis. This monomeric allows covalent attachment metathesis cofactors, and resulting capable catalyzing ring-closing Both genetic engineering were explored determine evolvability Additionally, exploration substrate revealed promising turnover numbers (>48) conversion rates (>96%).
